Change the input color format to computer RGB or YPbPr in the Color Settings On-Screen Display Menu (OSD). Select the player application (like media player) mute icon to mute or adjust the mute through the line out external speaker device.

Windows OS level computer volume change, line out (when you select echo canceling speakerphone) volume cannot be adjustable. You cannot use the mute on the connected device, which you should be able to when Echo Cancelling Speakerphone is selected.
